Electrochemical Synthesis of Composites Based on Silicon and Carbon Nanotube Compounds as a Platform for Biocatalyst Development ↗

Progress in the field of modern heterogeneous catalysts requires the development of composite materials with a well-developed surface area and sorption capacity, as well as the possibility of functionalization in a specific direction. This paper presents the results of electrochemical synthesis of composites based on an organosilica matrix and multi-walled carbon nanotubes for subsequent application as a platform for enzyme immobilization. By potentiostatic electrodeposition, composite coatings based on two types of organosilica sol (STPEG and STEG) were applied to the surface of a glassy carb…

Diagnostic Yield and Clinical Actionability of Genetic Testing in Neurodevelopmental Disorders: A Systematic Review and Meta-Analysis ↗

Genetic testing is increasingly central to the evaluation of neurodevelopmental disorders (NDDs), yet most evidence emphasises diagnostic yield rather than whether findings change clinical care. We conducted a systematic review and meta-analysis (Preferred Reporting Items for Systematic Reviews and Meta-Analyses [PRISMA] 2020) of genetic testing across the diagnostic pathway in NDD populations, focusing on autism spectrum disorder (ASD), intellectual disability, and global developmental delay, to quantify pooled diagnostic yield and clinical actionability. A systematic search of PubMed and Web…

CRISPR‐Scan: Cas13a Mismatch Tolerance Enabled Ultra‐Multiplexed Detection of Same‐Site Indels in Acute Myeloid Leukemia ↗

ABSTRACT Detection of gene mutations related to acute myeloid leukemia (AML) is crucial for its early diagnosis, personalized treatment, and recurrence monitoring. However, accurate detection of diverse mutations occurring at the same locus remains technically challenging. The CRISPR/Cas system, with its unique programmable nucleic acid recognition capabilities, provides a promising solution. Here, we elucidated that CRISPR/LwaCas13a is tolerant to consecutive substitutions but highly sensitive to insertions or deletions (indels) at specific positions of the spacer sequence, enabling rational…

Whole-genome sequencing reveals inter-household networks of gut-colonising ESBL-producing Escherichia coli in two rural Malawian districts ↗

Abstract Infection with extended-spectrum beta-lactamase-producing Escherichia coli (ESBL-Ec) is a global health concern that disproportionately affects sub-Saharan Africa. Gut mucosal colonisation is thought to precede invasive infection. Understanding ESBL-Ec colonisation and transmission across communities is therefore essential. Here, we investigate the genomic epidemiology and spatial structure of 159 gut-colonising ESBL-Ec isolates from the faeces of 211 people in two rural Malawian villages using longitudinal sampling (2023–24), whole-genome sequencing and household mapping. Colonisatio…

    PG-MTCDGNet: A Physics-Grounded Multi-Scale Time-Lagged Dynamic Graph Network for Structured Evidence Reporting in Planetary-Gearbox Fault Diagnosis ↗

Multichannel vibration from a planetary gearbox exhibits operating-condition-dependent interactions across both time scale and lag. Static undirected graphs cannot represent ordered temporal dependence, while repeated graph aggregation can attenuate localized impacts. This paper presents PG-MTCDGNet, a physics-grounded multi-scale time-lagged dynamic graph network for structured evidence reporting in planetary-gearbox fault diagnosis. Phase-synchronous X/Y vibration, transient residuals, Hilbert envelopes, cross-channel coupling, and encoder micro-speed are represented as eight physical nodes.…

PTM-PAL Pipeline: A Python Web Platform of Integrative Scoring System for Assessing the Functional Impact of Protein S-Palmitoylation ↗

S-palmitoylation is a reversible lipid modification that can alter protein localization, stability, interactions and signaling, but detection of a modified cysteine does not establish its functional importance. Post-translational Modification-Palmoylation Pipeline(PTM-PAL) is a web-accessible workflow for prioritizing candidate S-palmitoylation sites and modification configurations for functional follow-up. It integrates site-specific evolutionary context and UniProt annotation, paired wild-type and palmitoylated structural models, local protein-body solvent-accessible surface-area remodeling,…

HDAC Inhibitors Potentiate Doxorubicin Cytotoxicity in HepG2 Cells and Alter DNA-Repair- and p53-Associated Transcriptional Programs ↗

Histone deacetylase inhibitors (HDACis) can enhance responses to DNA-damaging chemotherapy, but how they alter doxorubicin (DOX)-induced transcriptional programs remains incompletely understood. We investigated the effects of valproic acid (VPA) and vorinostat (SAHA) on DOX cytotoxicity and associated molecular responses in HepG2 cells. HDAC inhibition, cell viability, apoptosis, and DOX IC50 values were assessed using histone H3 acetylation, cell counting, Alamar Blue assays, and cleaved caspase-3 immunofluorescence. Transcriptomic responses were analyzed by RNA sequencing, differential expre…

A high-quality telomere-to-telomere lumpy skin disease virus genome assembly ↗

Lumpy skin disease virus (LSDV) is an emerging livestock capripoxvirus (CaPV) that continues to cause substantial economic losses across Africa, Asia and Europe. However, important uncertainties remain regarding LSDV genome structure, particularly at the telomeric inverted terminal repeats (ITRs) that are central to host interaction, replication and adaptive evolution. Structural variation at the ITRs is driven by recombination during virus replication. Previous assemblies have relied primarily on short-read sequencing, which provides limited resolution of repeat-rich telomeric regions and pro…
