Earlier sources: Science
Science investigates the natural world through observation, experiments, and mathematical models. Explore papers and developments in biology, medicine, materials, neuroscience, physics, climate, and mathematics; a reported finding is not necessarily established consensus.
SYMTRI organizes observations from public sources into a living map. Connections reflect shared subject matter; they do not establish causation or endorse a source.
Open in the map ↗1374 classified signals in the fourteen-day window. Recent momentum: steady. These counts include signals awaiting readable evidence.
Earlier sources
Up to ten sources with readable evidence, ordered by their feed timestamp. These archive sources can be older than the live fourteen-day window. This list is a sample, not a measure of total activity. Dates distinguish paper publication, repository creation, model/dataset activity, Hacker News posts, and KEV catalog additions. “First archived” records when SYMTRI first stored the signal.

p38α MAPK integrates opposing oncogenic APC/β-catenin/TCF and GUCY2C tumor-suppressor axes in colorectal cancer ↗
Introduction: Sporadic colorectal cancers (CRCs) form following initiating APC mutations, inducing constitutive β-catenin/TCF signaling. Similarly, the guanylyl cyclase C (GUCY2C) tumor suppressor axis is universally silenced in CRC. While the GUCY2C receptor is retained, loss of its paracrine hormones guanylin (GUCA2A) and uroguanylin (GUCA2B) is among the earliest events in transformation. Mutant APC/β-catenin/TCF signaling driving CRC transcriptionally silences GUCY2C hormone expression while suppressing that oncogenic pathway reconstitutes hormone production opposing tumorigenesis. p38α op

Diabetes Polygenic Scores Predict Glycemic Indices and Insulin Use in Individuals with Atypical Diabetes ↗
Aims: /Hypothesis: Atypical forms of diabetes often provide diagnostic challenges and are suboptimally treated. We sought to determine whether type 1 and 2 diabetes polygenic scores (PS) would help improve classification of islet autoantibody-negative (IAb-) atypical diabetes cases. Methods: We implemented type 1 diabetes PS (T1D PS) and type 2 diabetes PS (T2D PS) in 309 IAb- individuals' genome sequencing from the Rare and Atypical Diabetes Network (RADIANT), the largest and most comprehensively phenotyped collection of unsolved and atypical diabetes cases. We performed regression analyses t

Establishment and characterization of LNCaP-TR-BoM, a bone metastasis-derived prostate cancer cell line with an intermediate AR-attenuated phenotype ↗
Background: Progression of prostate cancer under androgen receptor (AR) pathway-targeted therapy spans a phenotypic spectrum, from tumors that sustain AR pathway output to tumors that become independent of AR signaling. Experimental models that occupy the intermediate portion of this spectrum, in which AR is still expressed but pathway output is reduced, are comparatively scarce. Methods. LNCaP cells expressing a triple-modality reporter (LNCaP-TR) were injected into nude mice by the intracardiac route, and a bone lesion that arose in the middle spine of one animal was recovered and propagated

Hairpin Architecture and Extended Sequence Context Shape APOBEC3 Mutagenesis in Monkeypox Virus at Different Genomic Scales ↗
The 2022 global mpox outbreak was characterized by an unusual accumulation of more than fifty APOBEC-signature substitutions in MPXV genomes. A previous study suggested that these substitutions preferentially occur within and near hairpins or cruciforms - stem-loop structures formed by inverted repeats. Here, we show that this conclusion resulted from an inappropriate statistical analysis and, using two substantially larger datasets, demonstrate that APOBEC-signature mutations are specifically enriched at TC motifs located at the 3′ ends of hairpin loops, but not in other regions of hairpins/c

SLC25A28 is a synthetic lethal vulnerability in nucleotide excision repair-deficient cancers ↗
Approximately 10% of bladder, upper urinary tract, and endometrial cancers harbor deleterious somatic alterations in nucleotide excision repair (NER) pathway genes. Although platinum-based chemotherapy is the standard treatment for NER-deficient tumors, its utility is limited by nephrotoxicity, motivating alternative strategies that avoid direct DNA targeting. Here, we employed a genome-wide CRISPR-Cas9 screen to identify synthetic lethal vulnerabilities in NER-deficient cancer cells. An ERCC4-null RT112/84 bladder cancer cell line was generated and validated by Sanger sequencing and Western b

HIV-1 Ribonucleoprotein Complex Organization Facilitates Assembly of the Mature HIV-1 Capsid ↗
The encapsulation of the viral RNA condensate inside the capsid is a crucial step in the late stages of the HIV-1 replication cycle. While it is agreed that RNA encapsulation depends on RNA-bound integrase (IN), that capsid-bound IN-RNA filaments are present within the capsid, and that the ribonucleoprotein (RNP) complex takes up only a fraction of the capsid volume, the timing of filament formation and ribonucleoprotein (RNP) encapsulation are unclear. Here, we ran virion-scale, coarse grained molecular dynamics simulations to identify how the organization of the RNP and IN in particular affe

Inverse relationship between mitochondrial and non-coding nuclear DNA content in free-living kinetoplastids ↗
The evolution of genome size and eukaryotic genome organisation has puzzled evolutionary biologists for decades. Are differences in DNA content and protein-coding capacity of eukaryotic genomes a result of natural selection, or neutral processes? If they were shaped by natural selection, which features might be connected with these differences: cell volume, ecological strategy, availability of nutrients, etc.? Kinetoplastids are protists with compact nuclear genomes that almost entirely lack introns, but whose mitochondrial DNA content (kDNA) can rival that of their nuclei. We hypothesised tha

Polyphyletic origin and horizontal exchange drive the emergence of a novel resistance-breaking Fusarium oxysporum f. sp. lactucae race ↗
Fusarium oxysporum f. sp. lactucae (Fola), the cause of lettuce fusarium wilt, poses an increasing threat to global production due to the rapid emergence of resistance breaking isolates. However, the evolutionary origins and mechanisms underpinning this diversification remain poorly understood. We characterised 103 Fola isolates using molecular profiling, selecting ten key isolates for long-read whole-genome sequencing and performed in planta virulence assays for selected isolates. This included the first assemblies for Fola2 and Fola3, recent resistance-breaking variants and a highly divergen

Asp29Glu marks the re-humanised Mycobacterium tuberculosis complex lineages without affecting the protease melE (Rv2569c) ↗
The lineages of the Mycobacterium tuberculosis complex (MTBC) that returned to a human host after the animal radiation, L6 together with L9 and L10, share a small set of fixed variants. One of them, NC_000962.3:2894321:G:C (c.87C>G, p.Asp29Glu), falls in Rv2569c (melE), a macrophage-infection protease of the transglutaminase family described since 2004 and carrying the strongest in vivo phenotype signal; I asked whether it does anything. Across 142 673 genomes assigned to 705 clades, the variant is carried by 1400 of the 1402 genomes of the L6+L9+L10 node, the two exceptions being low-coverage

How to assess evidence about Science
Check each original source for its methods, release status, limitations, and supporting data. A repository announcement, a preprint, and a published study provide different kinds of evidence. SYMTRI’s subject classification helps discovery; it does not independently validate the source’s claims.
