Construction of red-spotted grouper nervous necrosis virus (RGNNV) nonstructural protein B1/B2 mutant strains and dynamics during infection. ↗

Nervous necrosis virus (NNV) is a neurotropic pathogen that causes serious losses in aquaculture worldwide. B1 and B2 proteins, nonstructural proteins of NNV, play vital roles in viral function. In this study, a plasmid-based reverse genetics system was established for red-spotted grouper nervous necrosis virus (RGNNV), and codon mutations on B1 and B2 proteins were introduced. One recombinant virus (recomb) and five mutant viruses (B1QM, B2QM, BoQM, B1JM, and B2JM) were successfully rescued. Mature virions of approximately 30 nm in diameter within intracellular vacuoles were observed under tr

Loss of Cotl1 disrupts mitochondrial fission and impairs learning and memory in mice. ↗

Mitochondrial morphology dynamics are essential for neuronal function and are regulated by fission and fusion machinery, in which the actin cytoskeleton plays a crucial role. Coactosin-like F-actin-binding protein 1 (Cotl1), a key component of the actin cytoskeleton, is gaining attention for its profound pathophysiological implications in human neurodegenerative disorders. However, its role in neuronal function remains unclear. Therefore, we aimed to explore the role of Cotl1 in mitochondrial morphology and neuronal dysfunction. We generated Cotl1-knockdown and Cotl1-K75E mutants to examine mi

Therapeutic potential of proline-rich peptide-1 (PRP-1) in counteracting amyloid-β-mediated cortical and hippocampal neurodegeneration in a rat model. ↗

Alzheimer's disease (AD) is associated with amyloid β (Aβ) accumulation, neuronal metabolic dysfunction, and structural degeneration. This study compares the neurotoxic effects of Aβ25-35 and Aβ1-42 in the rat cerebral cortex and hippocampus and evaluates the neuroprotective potential of proline rich peptide 1 (PRP-1). Amyloid peptides were administered bilaterally into the lateral ventricles, followed by intramuscular PRP-1 at 0.1 mg/kg once daily for three weeks, beginning 24 h after amyloid administration. Neuronal metabolic activity was assessed by Ca2⁺ dependent acid phosphatase histochem

Kidney-specific cell-free DNA as biomarker of kidney injury: Development and exploration of a multiplex digital droplet PCR assay. ↗

Cell-free DNA (cfDNA) is a promising non-invasive biomarker of organ injury. In transplantation, cfDNA is typically quantified by distinguishing donor- from recipient-derived DNA. However, we aimed to develop a multiplex digital droplet PCR (ddPCR) assay to measure kidney-derived cfDNA independent of donor-recipient differences as a marker of kidney injury. Five kidney-specific differentially methylated regions were identified in silico using The Cancer Genome Atlas and Gene Expression Omnibus data and combined into a multiplex ddPCR assay. The assay was validated in kidney implantation biopsi

Centrally administered SGLT2 inhibitor improves gut barrier function through the brain orexin, AMPK, dopamine D2 signaling and the vagal pathway in rats. ↗

Leaky gut, defined as increased intestinal permeability, has been associated with many diseases. Recent studies suggest the brain is involved in maintaining gut barrier function. Since tofogliflozin, a sodium/glucose cotransporter 2 (SGLT2) inhibitor acts centrally to increase food intake, and feeding and gut functions are deeply associated, this study aimed to clarify the hypothesis that gut barrier function is influenced by centrally administered SGLT2 inhibitors in rats using Evans blue method. Intracisternal SGLT2 inhibitor blocked colonic hyperpermeability in response to lipopolysaccharid

The role of social white matter tracts in susceptibility to phishing in aging. ↗

Vulnerability to fraud and financial exploitation among older adults is an increasing public health concern. Growing evidence supports that cognitive and socioemotional changes with age contribute to greater exploitation risk among older adults, while the neural mechanisms of heightened susceptibility to deception in aging are not well understood. Of note, white matter tracts are particularly impacted in aging and are likely implicated in deception detection. This study, for the first time, examined the extent to which age-related differences in white matter pathways subserving social cognitio
