THE INTERNET IS THINKING

Earlier sources: Science

Science investigates the natural world through observation, experiments, and mathematical models. Explore papers and developments in biology, medicine, materials, neuroscience, physics, climate, and mathematics; a reported finding is not necessarily established consensus.

SYMTRI organizes observations from public sources into a living map. Connections reflect shared subject matter; they do not establish causation or endorse a source.

Explainable pathomics and multi-omics define PDAC innervation subtypes linked to neuron–tumor crosstalk and immunotherapy response ↗

Neural regulation and an imbalanced immune microenvironment promote Pancreatic ductal adenocarcinoma (PDAC) progression. However, the molecular mechanisms and clinical implications of neuron–tumor interactions remain unclear. We integrated single-cell and spatial transcriptomic data to characterize neuron–tumor interactions in PDAC and classified patients into five innervation subtypes. We then assessed subtype differences in prognosis, immune microenvironment, and genomic instability. In parallel, we developed and externally validated a pathology-based machine learning model for subtype predi…

Clinical readiness of artificial intelligence for treat-to-target management in inflammatory bowel disease: from scores to decisions ↗

Artificial intelligence (AI) is increasingly used in inflammatory bowel disease (IBD), particularly for endoscopy, histology, transmural imaging, and prediction of treatment response. However, strong diagnostic or predictive performance does not necessarily confer clinical utility. This narrative review traces the development of AI in IBD from objective disease assessment and outcome prediction to therapeutic decision support within a treat-to-target framework. Endoscopic AI has the most mature evidence base, while applications in histology and transmural imaging are at earlier stages of clini…

From Brain to Blood to Skin: Compartment Biology, Interstitial Fluid Dynamics, and Microneedle Sampling of Neurological Biomarkers ↗

Proteins released after brain injury can be measured in blood, but a blood result does not directly measure the concentration at the site of injury. Between brain tissue and a peripheral sample, proteins may enter brain interstitial fluid (ISF), exchange with cerebrospinal fluid (CSF), leave through several drainage or barrier pathways, and undergo dilution and clearance. This narrative review examines those steps for glial fibrillary acidic protein (GFAP), ubiquitin C-terminal hydrolase-L1 (UCH-L1), neurofilament light chain (NfL), S100B, neuron-specific enolase (NSE), tau proteoforms, and am…

DuBCG: Unlocking In-Home, Clinically Comparable mmWave Cardiac Monitoring for Co-Sleepers via Physics-Informed Spatial Sensing ↗

Continuous, fine-grained cardiac monitoring during sleep is vital for cardiovascular health. Emerging as a promising alternative to cumbersome wearables, mmWave radar offers a contactless, privacy-preserving modality capable of capturing minute skin vibrations. However, transitioning mmWave radar from labs to real-world bedrooms remains challenging. Our rigorous empirical evaluation of state-of-the-art systems reveals a severe reliability gap: valid Inter-Beat Intervals (IBI) are captured for only 24%-42% of the night within an error threshold of 50 ms. Since clinical HRV analysis requires mil…

RuPHOTACs provide photocontrol over protein degradation with optimized properties for biological applications ↗

PROteolysis TArgeting Chimeras (PROTACs) are bifunctional molecules that catalyze degradation of selected proteins by inducing protein:protein interactions (PPIs) between E3 ubiquitin ligases and the protein of interest. A critical limitation is undesired effects in untargeted tissues, necessitating approaches to impose spatiotemporal control over PROTAC function. Here we present Ru(II) photocages that can be released with low energy light, providing triggered PROTAC activity on demand. The systems, termed Ruthenium-based PHOToActivated Chimeras (RuPHOTACs), were validated by targeting bromodo…

BRIP1, a G-quadruplex DNA helicase, protects against AID-induced genomic instability by limiting R-loop accumulation ↗

BRIP1 (BRCA1-interacting protein 1) or FANCJ is a 5′–3′ DNA helicase that plays critical roles in the maintenance of genome stability through its involvement in DNA repair, replication stress responses, and the resolution of DNA secondary structures or non-B DNAs. Defects in the BRIP1 helicase gene are associated with an increased risk of various cancers. It has been shown that BRIP1-deficient mice are predisposed to B-cell lymphoma. Because activation-induced cytidine deaminase (AID)-dependent genomic instability resulting from its mistargeting to non-immunoglobulin genes is a major hallmark…

Genome-Wide Identification of the PP2A Gene Family in Strawberry (Fragaria × ananassa) and Pharmacological Evidence for the Involvement of PP2A in ALA-Mediated Salt Tolerance ↗

Background: Protein phosphatase 2A (PP2A) is a conserved serine/threonine phosphatase that plays crucial roles in stress responses, yet its function in strawberry (Fragaria spp.) remains largely uncharacterized. This study aimed to characterize the PP2A gene family in octoploid strawberry and to investigate its involvement in ALA-induced salt tolerance. Methods: We identified PP2A genes in the octoploid strawberry ‘Benihoppe’ genome and classified them into scaffold (A), regulatory (B55, B56, B″), and catalytic (C) subunit families. Segmental duplication and selection pressure were analyzed us…

The Marb(e)lous BB84: Exploring Tangible Interfaces to Explain Quantum Key Distribution to Adults ↗

We explored the use of tangible interfaces to explain Quantum Key Distribution (QKD) - an inherently complex but increasingly important cybersecurity topic - to adults. Tangible interfaces can support the understanding of complex topics. Yet their role in cybersecurity remains underexplored, despite persistent demands for security awareness and Quantum Computing education. To address this, we applied a participatory approach: (1) exploring current explanations through a survey of QKD professionals ( N = 8) and analysis of popular YouTube videos, (2) conducting ideation workshops ( N = 9) to de…

DNA damage response associated miRNA dysregulation in Ataxia-telangiectasia ↗

BACKGROUND: Ataxia-telangiectasia (A-T) is a rare autosomal recessive genome-instability disorder due to pathogenic variants in ATM, a central kinase of the DNA double-strand break signaling, checkpoint activation and stress-response pathways. Although microRNAs (miRNAs) regulate DNA damage response (DDR), apoptosis, senescence and cell-cycle control, the stress-responsive behavior of selected ATM/DDR-associated miRNAs remains incompletely characterized in A-T fibroblast models. METHODS AND RESULTS: Four predefined ATM/DDR-associated candidate miRNAs (hsa-miR-34a-5p, hsa-miR-26a-5p, hsa-miR-10…
