Exploratory screening of wild-game tissues in Japan toward value-added utilization: estimated soluble protein/peptide yields and DPPH radical-scavenging and ACE-inhibitory activities after in vitro digestion ↗
Abstract This exploratory study aimed to compare estimated soluble protein/peptide yields and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging and angiotensin-converting enzyme (ACE)-inhibitory activities after in vitro digestion of eight tissues from wild-caught sika deer and wild boar and corresponding cattle and pig tissues, with relevance to value-added utilization in Japan. Eight tissues were obtained from sika deer and wild boar (n = 5 each) and commercially sourced cattle and pigs (n = 3 per tissue per species). Heated samples underwent gastric and intestinal digestion. DPPH and…

Pathogen genome enrichment sequencing to support a city-wide Legionnaires’ disease outbreak investigation ↗
Pathogen genomics can provide high-confidence source attribution for epidemiological investigations of Legionnaires' disease (LD) outbreaks. However, the utility of culture-based genomics can be undermined by long lead times for Legionella pneumophila culture or a lack of isolates when dealing with non-culturable bacteria or low-biomass specimens. Here, during a large 2024 LD outbreak in Melbourne, Australia, we compared direct specimen, hybridization-based enrichment sequencing with traditional culture-based whole-genome sequencing of 18 clinical and 5 environmental L. pneumophila specimens.…

Whole-Genome Analysis of Genomic Diversity and Antimicrobial Resistance Among Clinical Klebsiella pneumoniae Isolates from Farwaniya Hospital in Kuwait ↗
Background: K. pneumoniae is a major opportunistic pathogen recognized by the World Health Organization as a critical-priority antimicrobial-resistant pathogen. This study characterized isolates for antibiotic resistance determinants, virulence-associated genes, mobile genetic elements, and population structure of the isolates, and evaluated the correlation between antimicrobial resistance genotype and phenotype. Methods: Ninety-five isolates were collected from different clinical specimens in August–December 2024 in Farwaniya Hospital, Kuwait. Fifty-six isolates were selected for whole-genome…

Matrix conditioning and magnetic bead target transfer for fluorescence aptamer-CRISPR/Cas12a detection of kanamycin in river water ↗
Fluorescence aptamer-CRISPR/Cas12a assays provide efficient signal conversion and amplification for the detection of small molecules. However, their practical application in complex environmental water remains limited. Matrix components can cause signal loss before CRISPR amplification. In this study, a fluorescence aptamer-CRISPR/Cas12a assay was developed by integrating matrix conditioning and magnetic bead target transfer for kanamycin (KAN) detection in complex water matrices. Matrix conditioning reduced interference from humic acid (HA) and inorganic ions, while magnetic bead target trans…

Genome wide transcriptional changes underlie gradual and recurrent adaptation to protein malnutrition in zebrafish ↗
Lysosome-rich enterocytes (LREs) are a specialized population of intestinal cells that mediate the uptake and absorption of dietary proteins in fish and neonatal mammals. Loss of LRE function causes inhibited growth and poor survival due to protein malnutrition. Previously, we reported that in zebrafish loss of Plasmolipin (pllp), an endosomal membrane protein highly expressed in LREs, impairs LRE differentiation and dietary protein absorption, resulting in marked reduction in survival rates. Raising pllp homozygous mutants surviving to adulthood and in-crossing them for multiple generations r…

Identification of TMEM263 as an ER-resident protein that controls early stages of lipid droplet biogenesis ↗
Abstract Cell and organismal growth is controlled not only by the availability of nutrients, but also the ability to dynamically sense and respond to changes in metabolic demand. We have identified a protein, TMEM263 (also known as C12orf23) as a mechanistic link between growth and lipid metabolism. TMEM263 was discovered in a screen as an ER-resident protein, and we have characterised its role as both necessary and sufficient for lipid droplet formation. TMEM263 has two transmembrane domains that fold into a dynamic alpha hairpin which are essential for its localisation to the ER and to suppo…

Cotranslational membrane protein biogenesis by an EMC-bound translocon ↗
Secretory and membrane protein biogenesis occurs at ribosome-bound translocons in the endoplasmic reticulum (ER) membrane. Translocons consist of the Sec61 protein-conducting channel and various maturation factors including chaperones, modification enzymes and membrane insertases, which assemble dynamically in response to the biogenesis needs of a nascent polypeptide. In human cells, nascent multipass membrane proteins utilize the ER membrane protein complex (EMC) and the multipass translocon (MPT), but how these factors are deployed during biogenesis is unknown. Here we combine selective ribo…

3D GelMA-Printing of Pancreatic Endocrine Constructs via Magnetic Chitosan Nanoparticle-Mediated CRISPR/dCas9a Delivery: A Proof-of-Concept Study ↗
Diabetes mellitus affects nearly half a billion people worldwide and is primarily caused by the loss of insulin-producing cells, which are essential for maintaining normal blood glucose levels. In this study, we optimized a CRISPR activation (CRISPRa) system based on deactivated Cas9 (dCas9) to transfect HEK293 cells, which were subsequently differentiated into functional α-, β-, and δ-like cells as an engineering proof-of-concept. The CRISPRa system targeted multiple genes, including NGN3, NKX6.1 + MAFA, and PDX1, successfully inducing cell differentiation as confirmed by immunostaining. Thes…

How to assess evidence about Biotechnology
Check each original source for its methods, release status, limitations, and supporting data. A repository announcement, a preprint, and a published study provide different kinds of evidence. SYMTRI’s subject classification helps discovery; it does not independently validate the source’s claims.
