Phosphatidic Acid Vesicles Convert the Prion Protein into β-Sheet Fibrils with a Thermostable Structure but Temperature-Sensitive Packing ↗

Biomolecular cofactors, including nucleic acids and lipids, promote conversion of the cellular prion protein (PrPC) into pathogenic assemblies (prion scrapie, PrPSc), yet the mechanistic role of specific membrane lipids in the earliest stages remains unclear. Previous work showed that phosphatidic acid (PA) vesicles interact in vitro with murine recombinant PrP (recPrP) and trigger PrP aggregation without additional cofactors. Here, we dissect the PA-driven conversion pathway by monitoring PrP assembly and aggregate stability using spectroscopy and by comparing full-length recPrP with an N-ter…

Conformation‐Selective Binding Domains of Intramolecular Chemically Induced Protein Dimerisation Systems ↗

Chemically induced protein dimerisation (CID) is a widely used approach for detecting and actuating small-molecule-protein interactions. Here, we demonstrate that CID systems can be used as building blocks for higher-order ligand-controlled protein systems. To this end, we converted classical FK506- and cyclosporine A-controlled CIDs into single-chain pseudo-allosteric proteins. These systems displayed 15- and 50-fold gains in ligand affinity compared with the parental components operating in trans. Using these fusion proteins as baits in mRNA display, we identified FN3con binding domains that…

Genome-Wide Identification and Evolution Analysis of the bHLH Gene Family in Cucurbita pepo L. ↗

The basic helix–loop–helix (bHLH) gene family is one of the largest transcription factor families in plants and plays essential roles in growth, development, and responses to environmental stresses. However, the members and functional characteristics of the bHLH gene family in Cucurbita pepo L. remain largely unclear. In this study, a total of 175 bHLH genes were identified in Cucurbita pepo L., exhibiting considerable variation in protein length and structural features. Phylogenetic analysis classified these genes into 24 subgroups (Subgroup 1–24), with conserved motif compositions within eac…

Genome-Wide Identification and Expression Pattern Analysis of the Phenylalanine Ammonia-Lyase (PAL) Gene Family in Wheat Under Stripe Rust ↗

Phenylalanine ammonia-lyase (PAL) has been identified as the key rate-limiting enzyme of the phenylpropanoid metabolic pathway, which affects the synthesis of plant secondary metabolites and stress responses. To investigate the composition and function of the PAL gene family in wheat against wheat stripe rust, 54 TaPAL gene family members were identified based on the wheat IWGSC RefSeq v2.1 genome data. Phylogenetic analysis of PAL proteins from wheat, maize, rice, tobacco, and Arabidopsis indicated that wheat PAL genes are more closely related to those of maize and rice. Promoter analysis rev…

    Aging-associated SATB1 deficiency remodels 3D genome architecture and transcriptional programs in naive CD4 + T cells ↗

Precise three-dimensional (3D) genome organization is crucial for regulating gene expression during development, yet its role in age-related transcriptional changes and physiological decline remains elusive. Here, we show that aging reshapes chromatin architecture and gene regulation in murine naive CD4 + T cells, a quiescent T cell population essential for adaptive immunity. Aged naive CD4 + T cells exhibit intrinsic transcriptional reprogramming marked by increased expression of inflammatory and T cell activation–related genes, rendering them more prone to activation. Intriguingly, these cha…

Design and Evaluation of Novel Peptides Targeting Protein Disulfide Isomerase as Antiplatelet Agents ↗

Extracellular protein disulfide isomerase (PDI) is a critical mediator of thrombus formation and a promising target for novel antithrombotic therapies aiming to overcome the bleeding risks associated with current antiplatelet agents. This study aimed to design and evaluate novel, targeted peptide-based PDI inhibitors with a safe physiological profile. We utilized a bioinformatics-driven rational design approach, based on the evolutionary conservation of the PDI active site, to generate a library of 34 targeted peptides. The inhibitory mechanism and biological efficacy of the lead candidate, pe…

Protein Intake and Resistance Training Across Clinical Populations: A PRISMA-Guided Rapid Review of Observational and Randomized Evidence ↗

Background/Objectives: Guidance on protein and physical activity rests on population-wide minima with little differentiation by age or disease. We examined how protein intake and resistance training relate to muscle, function and mortality across clinical populations. Methods: PRISMA-guided rapid review: Fifty-eight logged PubMed queries returned 7544 unique records for screening (the analytic corpus); Europe PMC, Semantic Scholar and ClinicalTrials.gov were tested for coverage only. Estimates were extracted by a deterministic script and adjudicated by the authors; random-effects models were f…

Retinol-Binding Protein 4 in Obesity-Associated Metabolic Dysfunction: Mechanisms, Controversies, and Therapeutic Opportunities ↗

Retinol-binding protein 4 (RBP4) is the primary carrier of retinol in the circulation and is produced mainly by the liver. Interest in RBP4 expanded after reports of elevated circulating levels in insulin-resistant mice and some human populations, leading to its proposal as an adipokine, an immunometabolic mediator, and a biomarker of obesity-related metabolic disease. However, subsequent research has revealed that its biology is considerably more complex. The metabolic effects of RBP4 are influenced by multiple factors, including its tissue of origin, retinol-binding status, association with…

Genome-Wide Identification and Expression Dynamics of ABC Transporters in Morchella sextelata During Fruiting Body Development and Selenium Supplementation ↗

ATP-binding cassette (ABC) transporters constitute a superfamily of transmembrane transport proteins that are ubiquitously present across prokaryotic and eukaryotic organisms, exhibiting extensive substrate diversity and significant functional versatility. Yet, systematic genome-wide characterization of ABC transporters in Morchellasextelata remains largely unexplored. Here, a total of 33 ABC transporter genes were identified in M. sextelata and classified into eight subfamilies: ABCA (1), ABCB (7), ABCC (5), ABCD (6), ABCE (1), ABCF (4), ABCG (7), and ABCI (2), based on the features of domain…

Genome-Wide Identification and Functional Validation of the Cold-Tolerance WRKY Transcription Factor in Sweetpotato ↗

Sweetpotato (Ipomoea batatas (L.) Lam.) is a vital global food crop whose production and geographical distribution are severely constrained by low-temperature stress. While WRKY transcription factors are known to regulate plant stress responses, their specific roles in sweetpotato cold tolerance remain largely unexplored. In this study, we conducted a genome-wide identification of the WRKY family in the hexaploid sweetpotato genome, identifying 476 IbWRKYs classified into three major groups and seven subgroups. Synteny analyses revealed that whole-genome duplication (WGD)/segmental duplication…
